Saline-alkali soil severely reduces the productivity of crops,including maize(Zea mays).Although several genes associated with saline-alkali tolerance have been identified in maize,the underlying regulatory mechanism remains elusive.Here,we report a direct link between colonization by arbuscular mycorrhizal fungi(AMF)and saline-alkali tolerance in maize.We identify s75,a natural maize mutant that cannot survive under moderate saline-alkali soil conditions or establish AM symbioses.The saline-alkali hypersensitive phenotype of s75 is caused by a 1340-bp deletion in Zm00001d033915,designated as ZmL75.This gene encodes a glycerol-3-phosphate acyltransferase localized in the endoplasmic reticulum,and is responsible for AMF colonization.ZmL75 expression levels in roots correspond with the root length colonization(RLC)rate during early vegetative development.Notably,the s75 mutant line shows a complete loss of AMF colonization,along with alterations in the diversity and structure of its root fungal microbiota.Conversely,overexpression of ZmL75 increases the RLC rate and enhances tolerance to saline-alkali soil conditions.These results suggest that ZmL75 is required for symbiosis with AMF,which directly improves saline-alkali tolerance.Our findings provide insights into maize-AMF interactions and offer a potential strategy for maize improvement. 展开更多

Dental implants have restored masticatory function to over 100000000 individuals,yet almost 1000000 implants fail each year due to peri-implantitis,a disease triggered by peri-implant microbial dysbiosis.Our ability to prevent and treat peri-implantitis is hampered by a paucity of knowledge of how these biomes are acquired and the factors that engender normobiosis.Therefore,we combined a 3-month interventional study of 15 systemically and periodontally healthy adults with whole genome sequencing,finescale enumeration and graph theoretics to interrogate colonization dynamics in the pristine peri-implant sulcus.We discovered that colonization trajectories of implants differ substantially from adjoining teeth in acquisition of new members and development of functional synergies.Source-tracking algorithms revealed that this niche is initially seeded by bacteria trapped within the coverscrew chamber during implant placement.These pioneer species stably colonize the microbiome and exert a sustained influence on the ecosystem by serving as anchors of influential hubs and by providing functions that enable cell replication and biofilm maturation.Unlike the periodontal microbiome,recruitment of new members to the peri-implant community occurs on nepotistic principles.Maturation is accompanied by a progressive increase in anaerobiosis,however,the predominant functionalities are oxygen-dependent over the 12-weeks.The peri-implant community is easily perturbed following crown placement,but demonstrates remarkable resilience;returning to pre-perturbation states within three weeks.This study highlights important differences in the development of the periodontal and peri-implant ecosystems,and signposts the importance of placing implants in periodontally healthy individuals or following the successful resolution of periodontal disease. 展开更多

Colonization and development of the gut microbiome are crucial for the growth and health of calves.In this review,we summarized the colonization,beneficial nutrition,immune function of gut microbiota,function of the gut barrier,and the evolution of core microbiota in the gut of calves of different ages.Homeostasis of gut microbiome is beneficial for nutritional and immune system development of calves.Disruption of the gut microbiome leads to digestive diseases in calves,such as diarrhea and intestinal inflammation.Microbiota already exists in the gut of calf fetuses,and the colonization of microbiota continues to change dynamically under the influence of various factors,which include probiotics,diet,age,and genotype.Colonization depends on the interaction between the gut microbiota and the immune system of calves.The abundance and diversity of these commensal microbiota stabilize and play a critical role in the health of calves. 展开更多

Interactions between plants and soil microorganisms can influence the other interactions in which plants participate, including interactions with herbivores. Many fungi, including arbuscular mycorrhizal fungi(AMF), form symbiotic relationships with the roots they inhabit, and potentially alter defense against pests. The objective of this study was to document the extent of root colonization by AMF on non-flooded rice plants grown under conditions typical of commercial fields. We hypothesized that AMF naturally colonized rice plants in different rice producing field locations. Rice plant samples were collected from areas across the southern United States, including Texas, Mississippi, Arkansas and two research stations in Louisiana. We quantified the amount of AMF colonization in insecticide-free rice plants over three consecutive years(2014–2016). The results revealed natural colonization of AMF in all rice producing areas. In all the three years of survey, rice-AMF associations were the greatest in Arkansas followed by Mississippi and Texas. This research will help draw attention to natural colonization of AMF in rice producing areas that can impact future rice research and production by facilitating agricultural exploitation of the symbiosis. 展开更多

METHODS: The PCR cloning method identified the flgK gene. An isogenic flgK mutant was constructed by gene replacement and confirmed by Southern blot analysis and PCR analysis. The recombinant FlgK protein (r-FlgK) was purified. Electron microscopy (EM) was applied to demonstrate the flagella of H pylori. An in vitro motility test was assessed in semisolid medium. The densities of H pylori colonization with either the wild-type strain or its flgK mutant were compared among BALB/c mice with or without pre-immunization with r-FlgK. The serological responses to r-FlgK were analyzed for 70 clinical patients with different densities of H pylori colonization. RESULTS: From a duodenal ulcer strain, the flgK gene was cloned and it contained 1821 bp, with a 95.7% identity to the published sequences. No flagella were observed under EM for the mutant strain, which had a loss of motility. Hpylori density was lower in the BALB/c mice inoculated by the mutant or with pre-immunization with r-FlgK compared to unimmunized mice or mice inoculated by the wild-type strain (P 〈 0.05). In the H pylori-infected patients, the serological responses to r-FlgK were uniformly low in titer.CONCLUSION: FlgK encoded by flgK is important for flagella formation and H pylori motility. Deficiency in FlgK or an enhanced serological response to r-FlgK can interfere with Hpylori colonization. FlgK of Hpylori could be a novel target for vaccination. 展开更多

Helicobacter pylori(H.pylori)colonizes the stomach of humans and causes chronic infection.The majority of bacteria live in the mucus layer overlying the gastric epithelial cells and only a small proportion of bacteria are found interacting with the epithelial cells.The bacteria living in the gastric mucus may act as a reservoir of infection for the underlying cells which is essential for the development of disease.Colonization of gastric mucus is likely to be key to the establishment of chronic infection.How H.pylori manages to colonise and survive in the hostile environment of the human stomach and avoid removal by mucus flow and killing by gastric acid is the subject of this review.We also discuss how bacterial and host factors may together go some way to explaining the susceptibility to colonization and the outcome of infection in different individuals.H.pylori infection of the gastric mucosa has become a paradigm for chronic infection.Understanding of why H.pylori is such a successful pathogen may help us understand how other bacterial species colonise mucosal surfaces and cause disease. 展开更多

Oceanic islands are unique in their species composition,which is defined by arrival of colonizers via long distance dispersal followed by establishment of species followed in some cases by adaptive radiation.Evolutionary biologists identified traits facilitating successful colonization of islands as including polyploidy,self-compatibility, herbaceousness and ability for long-distance dispersal.Successful establishment and evolutionary diversification of lineages on islands often involves shifts to woodiness and shifts in methods of outcrossing as well as changes in dispersal ability.The genus Meflicope colonized numerous archipelagos throughout the Pacific including the Hawaiian Islands,where the lineage comprises currently 54endemic species and represents the largest radiation of woody plants on the islands.The wide distributional range of the genus illustrates its high dispersibility,most likely, due to adaption to bird dispersal.Here we investigate ploidy in the genus using flow cytometry and chromosome counting.We find the genus to be paleopolyploid with 2n =4x =36,a ploidy level characterizing the entire subfamily Amyridoideae and dating back to at least the Palaeocene.Therefore Hawaiian MeHcope have not undergone recent polyploidization prior to colonization of the islands.Thus Nlelicope retained colonization success while exhibiting a combination of traits that typically characterize well established island specialists while lacking some traits associated to successful colonizers. 展开更多

Arbuscular mycorrhizal fungi(AMF) and dark septate endophyte(DSE) colonization were investigated in three different plantation sites(Umdihar,Umsaw and Mawlein) of Meghalaya,northeast India.Isolation and identification of the AMF spore were conducted to evaluate the AMF diversity and host preference in terms of AMF species distribution and abundance in the plantation sites.Results showed that AMF colonization was significantly higher than dark septate endophyte colonization(p〉0.05).AMF and DSE colonization had a narrow range of colonization,varying from 50.91%-58.95% and 1.84%-4.11%,respectively.Spore density varied significantly in all the sites(p〉0.05).Out of 29 species identified from 7 genera,the species from Glomus was found to be highly abundant.Sorenson coefficient(Cs) ranged from 0.35-7.0.Species richness varied from 2.0-2.9 in the sites.Total species richness was significantly correlated with total relative abundance(p=0.001).The distribution,abundance and principal component analysis plot suggest that Glomus macrocarpum,G.multicaulis,G.constrictum and Acaulospora sp 1 were the most host preferred species which possibly may favour the host with proper nutrient acquisition and growth. 展开更多

Biofilm-producing bacteria can decrease Cd uptake in vegetables, but mechanisms underlying this effect are poorly characterized. In this study, two mutant strains B12ΔYwcc and B12ΔSlr R were constructed from a biofilm-producing Bacillus subtilis strain B12. Then, the impacts of strain B12 and its high biofilm-producing mutant strain B12ΔYwcc and low biofilmproducing mutant strain B12ΔSlr R on Cd availability and uptake in Chinese cabbage and the related mechanisms were investigated in the Cd-polluted soil. Strain B12 and its mutants B12ΔYwcc and B12ΔSlr R increased the dry biomasses of edible tissues by 54%–130% compared with the controls. Strain B12 and its mutant B12ΔYwcc reduced the soil available Cd content by 36%–50% and root and edible tissue Cd contents by 23%–50% compared with the controls. Furthermore, the mutant strain B12ΔYwcc reduced the edible tissue Cd content by40% and increased the polysaccharide content by 23%, invertase activity by 139%, and gene copies of the cum A by 4.5-fold, eps A by 7.1-fold, and cad A by 4.3-fold, which were involved in Cd adsorption in the rhizosphere soils, respectively, compared with strain B12. The polysaccharide content and cum A, eps A, and cad A gene copy numbers showed significantly reverse correlations with the available Cd content. Notably, the mutant strain B12ΔYwcc showed better ability to colonize the vegetable root surface than strain B12. These findings demonstrated that the biofilm-overproducing mutant strain B12ΔYwcc increased the polysaccharide production and Cd-immobilizing related cum A, eps A, and cad A gene copies, resulting in lower Cd availability and accumulation in Chinese cabbage in the Cd-polluted soil. 展开更多

The origin and colonization history of Kurixalus,a genus of small arboreal tree frogs breeding exclusively in shallow swamps,is under disputed.On the basis of comprehensive sampling program,the evolutionary history of Kurixalus is investigated based on 3 mitochondrial genes.Our results indicate that the genus Kurixalus originated in the Asian mainland and subsequently arrived at its current distribution in Borneo,Taiwan,Ryukyu,and Hainan islands by a series of dispersal events.Moreover,the colonization of Taiwan from mainland Asia has occurred 2 times.The initial colonization of Taiwan occurred at 3.46-8.68 Mya(95%highest posterior density),which rejects the hypothesis that Kurixalus probably originated from Taiwan during the early Oligocene and favors the model of Neogene-origin rather than the model of Quaternary-origin for Taiwan Residents Kurixalus.Kurixalus eiffingeri has dispersed from Taiwan to the Ryukyus once or 2 times pending more data.Both transoceanic dispersal and landbridge dispersal have played a role in the colonization process;the former resulted in the colonization of Taiwan and the Ryukyus and the latter led to the colonizati on of Bor neo and Hain an. 展开更多

Sclerotinia stem rot,caused by Sclerotinia sclerotiorum,is a destructive soil-borne disease leading to huge yield loss.We previously reported that Klebsiella variicola FH-1 could degrade atrazine herbicides,and the vegetative growth of atrazine-sensitive crops(i.e.,soybean)was significantly increased in the FH-1-treated soil.Interestingly,we found that FH-1 could promote soybean growth and induce resistance to S.sclerotiorum.In our study,strain FH-1 could grow in a nitrogen-free environment,dissolve inorganic phosphorus and potassium,and produce indoleacetic acid and a siderophore.The results of pot experiments showed that K.variicola FH-1 promoted soybean plant development,substantially improving plant height,fresh weight,and root length,and induced resistance against S.sclerotiorum infection in soybean leaves.The area under the disease progression curve(AUDPC)for treatment with strain FH-1 was significantly lower than the control and was reduced by up to 42.2%within 48 h(P<0.001).Moreover,strain FH-1 rcovered the activities of catalase,superoxide dismutase,peroxidase,phenylalanine ammonia lyase,and polyphenol oxidase,which are involved in plant protection,and reduced malondialdehyde accumulation in the leaves.The mechanism of induction of resistance appeared to be primarily resulted from the enhancement of transcript levels of PR10,PR12,AOS,CHS,and PDF1.2 genes.The colonization of FH-1 on soybean root,determined using CLSM and SEM,revealed that FH-1 colonized soybean root surfaces,root hairs,and exodermis to form biofilms.In summary,K.variicola FH-1 exhibited the biological control potential by inducing resistance in soybean against S.sclerotiorum infection,providing new suggestions for green prevention and control. 展开更多

The present study evaluated the effect of non-thermal plasma on skin wound healing in BalB/c mice.Two 6-mm wounds along the both sides of the spine were created on the back of each mouse(n=80) by using a punch biopsy.The mice were assigned randomly into two groups,with 40 animals in each group:a non-thermal plasma group in which the mice were treated with the non-thermal plasma;a control group in which the mice were left to heal naturally.Wound healing was evaluated on postoperative days(POD) 4,7,10 and 14(n=5 per group in each POD) by percentage of wound closure.The mice was euthanized on POD 1,4,7,10,14,21,28 and 35(n=1 in each POD).The wounds were removed,routinely fixed,paraffin-embedded,sectioned and HE-stained.A modified scoring system was used to evaluate the wounds.The results showed that acute inflammation peaked on POD 4 in non-thermal plasma group,earlier than in control group in which acute inflammation reached a peak on POD 7,and the acute inflammation scores were much lower in non-thermal group than in control group on POD 7(P0.05).The amount of granular tissue was greater on POD 4 and 7 in non-thermal group than in control group(P0.05).The re-epithelialization score and the neovasularization score were increased significantly in non-thermal group when compared with control group on POD 7 and 10(P0.05 for all).The count of bacterial colonies was 103 CFU/mL on POD 4 and 20 CFU/mL on POD 7,significantly lower than that in control group(109 CFU/mL on POD 4 and 1012 CFU/mL on the POD 7)(P0.05).It was suggested that the non-thermal plasma facilitates the wound healing by suppressing bacterial colo-nization. 展开更多

Viable cell count was used to determine whether Metschnikowia sp.C14 can colonize the intestine of juvenile sea cucumber Apostichopus japonicus.Sea cucumber individuals were divided into two groups,which were fed the control diet for 38 days or the C14-supplemented diet at 105 cells g−1 diet for 28 days,then the control diet from day 29 to day 38.The number of C14 cells in the intestine of sea cucumber fed the C14-supplemented diet significantly increased from day 7 to day 28,and decreased from day 29 to day 38.Sea cucumber fed with the diet containing C14 showed a significant increase in trypsin activity and lipase activity from day 21 to day 33 compared with the control.Feeding C14 significantly improved the phagocytic activity and respiratory burst in coelomocytes from day 21 to day 35 and from day 14 to day 38,respectively.In addition,there was an obvious enhancement in lysozyme activity(from day 21 to day 38 or day 33),phenoloxidase activity(from day 21 to day 28)and total nitric oxide synthase activity(from day 14 to day 38)in coelomic fluid supernatant and/or coelomocyte cell lysate supernatant compared with the control.There were significant positive correlations between the number of C14 cells colonizing the intestine and trypsin activity of the intestine,lysozyme activity of the coelomic fluid supernatant and coelomocyte lysate supernatant from sea cucumber.These data suggested that the number of C14 cells should be maintained at 105 cfu(colony-forming units)g−1 intestine material for the maximum benefit. 展开更多

Fragmentation and loss of habitats due to natural disasters, like earthquakes and earthquaketriggered debris flows are existing threats to the long- term survival of the giant panda (Ailuropoda melanoleuca). To better understand natural recovery processes of the damaged habitat, field investigation and laboratory analysis were used to analyze relationships between plant colonization and soil characteristics in an over 3o-year natural recovery of a damaged giant panda habitat in a debris flow gully after the 1976 Songpan-Pingwu earthquake in Sichuan Province, China. Four different damaged sites were selected that located at the center of the gully (center), on a flat alluvial fan (fan), in a side slope of the gully (slope), and at the ecotone between the gully and native forest (ecotone). Vegetation characteristics, soil physicochemical properties, and microbial biomass in the different sites and soil depths were measured. After the natural recovery, the soil fertility, water retention, and microbial biomass were highest at ecotone, followed by fan, slope, and center. Only a few perennial herbs colonized at center; shrubs started to invade at fan and slope, and the native trees dominated the community of ecotone. Furthermore, Fargesia spathacea (food for the giant panda) started to be re-established at ecotone, and the community characteristic of ecotone recovered similarly to the native habitat. These results suggested that improving the soil fertility, water retaining capacity and microbial biomass is fundamental to the plant colonization, particular for F. spathacea's re- establishment in a damaged giant panda habitat. 展开更多

To understand arbuscular mycorrhizal(AM)fungi resources and develop AM fungal species in ornamental plants with saline-alkaline tolerances,Iris lactea,which grows in the Songnen saline-alkaline grassland with a high ornamental value,was selected as the experimental material,and the colonization characteristics of its roots and the AM fungal diversity in its rhizosphere were explored.The results of the observations and calculations of mycorrhizae from ten different samples showed that AM fungi colonized the roots of I.lactea and formed Arum-type mycorrhizal structures.There was a significant correlation between soil spore density and pH value,while the colonization rate showed a fluctuating trend with increasing pH values.The observed colonization intensities were of Levels II(1%–10%)or III(11%–50%),and the vesicle abundances were of grades A2 or A3 among different sites.AM fungi produced a large number of mycelia and vesicles in the roots of I.lactea after colonization.Thirty-seven species belonging to 15 genera of AM fungi were isolated from the rhizosphere of I.lactea and identified by morphological identification.Funneliformis and Glomus were the dominant genera,accounting for 21.79%and 20.85%of the total number,respectively.F.mosseae and Rhizophagus intraradices were isolated in all samples with importance values of 58.62 and 51.19,respectively.These results are expected to provide a theoretical basis for the analysis of the salt tolerance mechanism of I.lactea and for the discovery,exploration and further screening of AM fungal resources with salinity tolerances in saline-alkaline soils. 展开更多

Roots and rhizosphere soils of Albizia procera Benth., Capsicum frutescens L, Curcuma domestica Vahl., Dalbergia sissoo Roxb. and Swietenia macrophylla King. from Dashmail; C. domestica, D. sissoo, Eucalyptus camaldulensis Dehnn., Gmelina arborea (Roxb) DC and Oryza sativa L. from Kantaji and C. domestica, D. sissoo, Litchi chinensis Sonn. and O. sativa from Ramsagar were collected. Roots and soils were assessed following standard methods. The range of AM colonization was recorded 36%-79% from Dashmail. The highest AM colonization was recorded in C. frutescens (79%) and the lowest was in C. domestica (36%). The range of colonization was recorded as 33%-70% from Kantaji. The highest AM colonization was recorded in G. arborea (70%) and the lowest was in O. sativa (33%). The range of AM colonization was recorded as 35%-70% from Ramsagar. The highest AM colonization was recorded in D. sissoo (70%) and the lowest was in O. sativa (35%). Arbuscular mycorrhizal spore population varied from 54 to 140/100g dry soil in the soils from Dashmail. The highest was in the soils of D. sissoo (140) and the lowest was in C. domestica (54). The spore population varied from 63 to 221 in Kantaji. The highest was in G. arborea (221) and the lowest was in O. sativa (63). The range population in Ramsagar varied from 69 to 160. The highest was recorded in D. sissoo (160) and the lowest was in L. chinensis (69). No significant relationship of soil pH and soil OM with AM colonization and with spore population was observed. Simpson's index of diversity (Ds) and Shannon's index of diversity- (Hs) were highest in the soil of D. sissoo from Kantaji and the lowest in the soils of O. sativa from Ramsagar. Biodiversity of AM colonization, spore population and the distribution of AM fungi in the rhizosphere soils of different agroforestry plants indicated the occurrence of AM fungi, mycotrophic nature of the trees and crop species, contribution and necessity of AM fungi and the AM dependence of the agroforestry plants growing in Dinajpur district of Bangladesh. 展开更多

To investigate the bacterial colonization on zirconium oxide and titanium surfaces in vivo quantitatively using a confocal laser scanning microscope (CLSM). Ten samples of zirconium oxide ceramic and commercially pure titanium were fabricated and polished using silicon carbide abrasive paper. One sample from each group was evaluated topographic pattern under a scanning electron microscope. One sample from each group was to evaluate roughness using a profilometer. Eight volunteers were selected. The samples were cemented at the buccal surfaces of upper first molars. All samples were removed after 48 hours, immersed in SYTO-9 and propidium iodide fluorescent to stain for adherent bacteria and obseIved with CLSM. Fewer bacteria were observed in zirconia group than titanium group. However, there was no statistical difference between two groups. The experimental results demonstrate that zirconium oxide may be considered as a promising material for dental implant abutments. 展开更多

AIM:To investigate the effect of celecoxib,a selective COX-2 inhibitor,on Helicobacter pylori(H.pylori) colonization-related factors and its mechanism.METHODS:After co-incubation with celecoxib,morphology of H.pylori strain 26695 was observed under a transmission electron microscope.Flagella motility was assessed by stab agar motility test.Adherence of H.pylori to AGS cells was determined by enzyme linked immunosorbent assay.Levels of mRNA expression in flagellar genes(flaA,flaB),urease genes(ureA,ureB)and adhesin genes(babA,sabA,alpA,alpB,hpaA,hopZ)were measured by real-time polymerase chain reaction.RESULTS:Separation and non-integrity of bacterial cell wall,rarefaction and asymmetry of cytoplasm,and even lysis of H.pylori were observed in the presence of celecoxib.When H.pylori strains were incubated in the presence of celecoxib,their flagellar motility and adherence to AGS cells were inhibited.The expression of ureA,ureB,babA,sabA,alpA,alpB,hpaA,hopZ was up-regulated while the expression of flaA,flaB was down-regulated in the presence of celecoxib.CONCLUSION:Celecoxib inhibits flagellar motility and adherence of H.pylori to AGS cells,and destructs their normal structure in vitro. 展开更多
