Neuroscience and neurology research is dominated by experimentation with rodents.Around 75%of neurology disease-associated genes have orthologs in Drosophila mel-anogaster,the fruit fly amenable to complex neurologica...Neuroscience and neurology research is dominated by experimentation with rodents.Around 75%of neurology disease-associated genes have orthologs in Drosophila mel-anogaster,the fruit fly amenable to complex neurological and behavioral investiga-tions.However,non-vertebrate models including Drosophila have so far been unable to significantly replace mice and rats in this field of studies.One reason for this situ-ation is the predominance of gene overexpression(and gene loss-of-function)meth-odologies used when establishing a Drosophila model of a given neurological disease,a strategy that does not recapitulate accurately enough the genetic disease condi-tions.I argue here the need for a systematic humanization approach,whereby the Drosophila orthologs of human disease genes are replaced with the human sequences.This approach will identify the list of diseases and the underlying genes that can be adequately modeled in the fruit fly.I discuss the neurological disease genes to which this systematic humanization approach should be applied and provide an example of such an application,and consider its importance for subsequent disease modeling and drug discovery in Drosophila.I argue that this paradigm will not only advance our un-derstanding of the molecular etiology of a number of neurological disorders,but will also gradually enable researchers to reduce experimentation using rodent models of multiple neurological diseases and eventually replace these models.展开更多
Shortening working time and making it more flexible, which have been observed in Europe since mid-20th century, are perceived as the main features and conditions for labour humanization as well as the sign of social p...Shortening working time and making it more flexible, which have been observed in Europe since mid-20th century, are perceived as the main features and conditions for labour humanization as well as the sign of social progress. The aim of the article has been showing negative consequences of this phenomenon, especially for maintaining balance between work and private life. It was based on hypothesis that the tendency to shorten the working time and to make it more flexible cannot be seen only as a factor of humanization, as the achievements of the social policy in Europe have been accompanied by many negative aspects, having a negative influence over both employees and employers. One of those observed occurrences were progressive disturbances in balancing work and private life. The considerations have been based on critical literature analysis method, the analysis of formal legal documents and statistics reports and sources. The transformations of working time and their influence over free time shortage and thus the imbalance between work and private life have been analysed. The theoretical-problem analyses allowed to reach the conclusion that the transformations of working time, related to negative changes in its organization, can be perceived as a factor of dehumanization of one's work.展开更多

The brain is the most complex human organ,and commonly used models,such as two-dimensional-cell cultures and animal brains,often lack the sophistication needed to accurately use in research.In this context,human cereb...The brain is the most complex human organ,and commonly used models,such as two-dimensional-cell cultures and animal brains,often lack the sophistication needed to accurately use in research.In this context,human cerebral organoids have emerged as valuable tools offering a more complex,versatile,and human-relevant system than traditional animal models,which are often unable to replicate the intricate architecture and functionality of the human brain.Since human cerebral organoids are a state-of-the-art model for the study of neurodevelopment and different pathologies affecting the brain,this field is currently under constant development,and work in this area is abundant.In this review,we give a complete overview of human cerebral organoids technology,starting from the different types of protocols that exist to generate different human cerebral organoids.We continue with the use of brain organoids for the study of brain pathologies,highlighting neurodevelopmental,psychiatric,neurodegenerative,brain tumor,and infectious diseases.Because of the potential value of human cerebral organoids,we describe their use in transplantation,drug screening,and toxicology assays.We also discuss the technologies available to study cell diversity and physiological characteristics of organoids.Finally,we summarize the limitations that currently exist in the field,such as the development of vasculature and microglia,and highlight some of the novel approaches being pursued through bioengineering.展开更多
Background Regular physical training induces adaptive effects across multiple organ systems,highlighting the existence of inter-organ communication networks.However,the molecular mechanisms underlying both exercise-in...Background Regular physical training induces adaptive effects across multiple organ systems,highlighting the existence of inter-organ communication networks.However,the molecular mechanisms underlying both exercise-induced adaptations and organ-to-organ signaling are not fully characterized.Circulating extracellular vesicles(EVs),including exosomes,carry molecules like microRNAs(miRNAs)that may mediate tissue crosstalk.This study aimed to identify specific exercise training-responsive miRNAs that affect skeletal muscle function.Methods miRNA expression profiles of serum-derived EVs were analyzed in healthy young individuals before and after 3 weeks endurance exercise training.Exercise training-responsive miRNAs were then validated for a functional role in cellular metabolic processes in human myotubes.Results We identified several exercise training-responsive miRNAs within exosome-rich EVs in serum,including miR-136-3p.In human myotubes,miR-136-3p enhanced glucose uptake and targeted the nardilysin convertase(NRDC)gene.Transfection of miR-136-3p or silencing of NRDC induced a shift towards glycolytic metabolism in mitochondria and modulated gene expressions related to myogenesis.Pancreatic islets were identified as a potential source of miR-136-3p based on in silico analysis of gene expression and a molecular analysis of conditioned media from isolated pancreatic islets.Conclusion MiR-136-3p is an endurance training-responsive molecular transducer that modulates glucose metabolism and cellular proliferation in myocytes.Associated with EVs,extracellular miR-136-3p may serve as a molecular messenger to communicate islet–skeletal muscle crosstalk after exercise.Extracellular miR-136-3p may serve as a molecular messenger to communicate islet–skeletal muscle crosstalk.Our results highlight a miRNA-mediated mechanism that participates in inter-organ communication to fine tune the metabolic adaptations to exercise.展开更多

Traditional targeted analyses often overlook unknown or emerging contaminants,highlighting the significance of nontarget and suspect screening approaches.A novel and high-sensitivity methodology for nontarget analysis...Traditional targeted analyses often overlook unknown or emerging contaminants,highlighting the significance of nontarget and suspect screening approaches.A novel and high-sensitivity methodology for nontarget analysis of organic pollutants in human serum was newly-developed based on gas chromatography coupled with quadrupole time-of-flight high-resolution mass spectrometry.The extraction protocol employing an acetonitrile-ethyl acetate(9:1,V:V)mixture significantly improved the extraction efficiency while minimizing matrix effect.A hybridized analytical strategy integrating nontarget and suspect screening was developed to achieve comprehensive identification and classification of pollutants,employing the National Institute of Standards and Technology(NIST)20 library and Agilent Technologies Personal Compound Database and Library(PCDL).This approach successfully characterized 273 organic contaminants spanning 12 categories,including polycyclic aromatic hydrocarbons(PAHs)and their derivatives,esters,and phenolic compounds in human serum,with a significant increase in detection specificity compared to conventional workflows.The methodology used serum samples of the workers from coking industry,revealing widespread contamination dominated by PAHs and PAH derivatives.Among the target analytes,three were identified solely by NIST and six solely by PCDL,indicating the complementary benefits of combining these different databases.Notably,this work reported the first confirmed detection of 2-naphthalenamine in human serum.This optimized approach demonstrates enhanced sensitivity and reliability in serum analysis,advancing biomonitoring capabilities and providing a deep understanding of human exposure to environmental pollutants.展开更多
Understanding the impacts of human activities on the plateau’s living environment is essential for advancing modernization pathways that promote harmony between humanity and nature.However,studies on the dynamic inte...Understanding the impacts of human activities on the plateau’s living environment is essential for advancing modernization pathways that promote harmony between humanity and nature.However,studies on the dynamic interactions between human activities and the living environment on the Qinghai-Xizang Plateau(QXP)remain limited,with a paucity of quantitative relationship analyses.This study established an assessment framework to evaluate human influences on the living environment in QXP,using data on typical human activities,ecological conditions,and human settlements.Within this framework,the spatial analysis methods and the coupling coordination model were used to examine the spatio-temporal characteristics and relationship of human activities and living environment on the QXP from 2000 to 2020.The geographical detector model was then applied to identify the key factors influencing the plateau’s human living environment.Subsequently,the four-quadrant analysis model was adopted to assess human influences on the living environment.The results indicate that the human activity intensity(HAI)on the QXP remained relatively low yet increased by 15.41%from 2000 to 2020.Spatially,the human living environment quality(LEQ)improved from northwest to southeast,with 61.14%of the areas remaining stable and 18.47%experiencing slight improvement.The analysis of coupling coordination revealed a continuous improvement between the HAI and LEQ,with the areas of high and relatively high coordinated types increasing by more than 9%.Precipitation and urban-rural construction were identified as the primary factors influencing changes in the LEQ.The interaction between the HAI and LEQ was strengthening,with 40.44%classified as coordinated development type and 38.35%as development-environment conflict type.These findings provide valuable insights for enhancing the resilience of human settlements and promoting green development across the plateau.展开更多

Neonicotinoid insecticides(NEOs)are a group of widely used insecticides in agriculture,livestock,and pest control for pets.However,increasing reports on their adverse effects on non-target organisms have aroused growi...Neonicotinoid insecticides(NEOs)are a group of widely used insecticides in agriculture,livestock,and pest control for pets.However,increasing reports on their adverse effects on non-target organisms have aroused growing public concerns.Therefore,it is necessary to evaluate NEO exposure and the related human health.This review summarized the human biomonitoring studies on NEOs,and the epidemiological,in vitro,and in vivo toxicological investigations on the health effects of NEOs.Human biomonitoring studies revealed significant changes in NEO detection frequencies,concentrations,and profiles across different human populations,suggesting potential variances in exposure patterns or metabolic processes.Certain metabolites may be more sensitive biomarkers than their parent molecules,as evidenced by higher levels of some NEO metabolites than those of their parent compounds.Although human epidemiological data has revealed associations between NEO exposure and diverse detrimental health outcomes including reproductive and developmental abnormalities,oxidative damage,etc.,the cross-sectional design of most studies cannot elucidate causality,necessitating further prospective research.In vitro and in vivo studies have shown various toxic effects of NEOs,including neurotoxicity,hepatotoxicity,reproductive toxicity,genotoxicity,etc.Further research on the molecular mechanisms beyond oxidative stress is worthwhile.Given metabolic activation,additional metabolites of both conventional and new generation NEOs as well as NEO-like compounds need to be determined,and their toxicities should be evaluated.This review can be contributed to the comprehensive understanding of the threat from extensive exposure to NEOs and proposed future research directions can aid to improve studies on health effects of NEOs.展开更多

In this short novel, the protagonist Buck is humanized by the author. He is endowed with lots of human qualities, such as loyalty, brave, self-esteem, intelligence and so on. What are the reasons of humanization of the dog protagonist.? Finding out the reasons could help us understand this novel better and it may offer some references for the research on Jack London' s animal novels.

People have higher and higher requirements for living environment, space and layout, especially in todays low-carbon society. People prefer a healthy, happy and comfortable living environment. With the continuous improvement of peoples living needs, the design concept of "people-oriented" in architectural design also appears. It adopts humanized design to create a warm and pleasant atmosphere. Therefore, in the current residential building design, it is necessary to meet the needs of residents. While living needs, we should also consider the impact of the living environment on all aspects of human beings, so as to create a livable living environment for human beings.

摘要The brain is the most complex human organ,and commonly used models,such as two-dimensional-cell cultures and animal brains,often lack the sophistication needed to accurately use in research.In this context,human cerebral organoids have emerged as valuable tools offering a more complex,versatile,and human-relevant system than traditional animal models,which are often unable to replicate the intricate architecture and functionality of the human brain.Since human cerebral organoids are a state-of-the-art model for the study of neurodevelopment and different pathologies affecting the brain,this field is currently under constant development,and work in this area is abundant.In this review,we give a complete overview of human cerebral organoids technology,starting from the different types of protocols that exist to generate different human cerebral organoids.We continue with the use of brain organoids for the study of brain pathologies,highlighting neurodevelopmental,psychiatric,neurodegenerative,brain tumor,and infectious diseases.Because of the potential value of human cerebral organoids,we describe their use in transplantation,drug screening,and toxicology assays.We also discuss the technologies available to study cell diversity and physiological characteristics of organoids.Finally,we summarize the limitations that currently exist in the field,such as the development of vasculature and microglia,and highlight some of the novel approaches being pursued through bioengineering.

摘要Background Regular physical training induces adaptive effects across multiple organ systems,highlighting the existence of inter-organ communication networks.However,the molecular mechanisms underlying both exercise-induced adaptations and organ-to-organ signaling are not fully characterized.Circulating extracellular vesicles(EVs),including exosomes,carry molecules like microRNAs(miRNAs)that may mediate tissue crosstalk.This study aimed to identify specific exercise training-responsive miRNAs that affect skeletal muscle function.Methods miRNA expression profiles of serum-derived EVs were analyzed in healthy young individuals before and after 3 weeks endurance exercise training.Exercise training-responsive miRNAs were then validated for a functional role in cellular metabolic processes in human myotubes.Results We identified several exercise training-responsive miRNAs within exosome-rich EVs in serum,including miR-136-3p.In human myotubes,miR-136-3p enhanced glucose uptake and targeted the nardilysin convertase(NRDC)gene.Transfection of miR-136-3p or silencing of NRDC induced a shift towards glycolytic metabolism in mitochondria and modulated gene expressions related to myogenesis.Pancreatic islets were identified as a potential source of miR-136-3p based on in silico analysis of gene expression and a molecular analysis of conditioned media from isolated pancreatic islets.Conclusion MiR-136-3p is an endurance training-responsive molecular transducer that modulates glucose metabolism and cellular proliferation in myocytes.Associated with EVs,extracellular miR-136-3p may serve as a molecular messenger to communicate islet–skeletal muscle crosstalk after exercise.Extracellular miR-136-3p may serve as a molecular messenger to communicate islet–skeletal muscle crosstalk.Our results highlight a miRNA-mediated mechanism that participates in inter-organ communication to fine tune the metabolic adaptations to exercise.

摘要Resistant starch(RS)comprises starch fractions that resist digestion in the small intestine and reach the colon,where they are fermented by the microbiota.Resistant starch harbors functional properties and healthpromoting ingredients that can regulate blood glucose and lipid levels,prevent cancer,and enhance the quality of life.Consequently,new technologies for the preparation of RS are continually being developed to support its industrial production.This review describes the structural and nutritional properties of RS and examines recent advancements in RS preparation methods.Emphasis is placed on how RS structure influences its properties and the physiological mechanisms in vivo.This review aims to stimulate further research into the preparation methods,functional characteristics,and utilization of RS,thereby supporting ongoing developments in the food industry.

摘要Traditional targeted analyses often overlook unknown or emerging contaminants,highlighting the significance of nontarget and suspect screening approaches.A novel and high-sensitivity methodology for nontarget analysis of organic pollutants in human serum was newly-developed based on gas chromatography coupled with quadrupole time-of-flight high-resolution mass spectrometry.The extraction protocol employing an acetonitrile-ethyl acetate(9:1,V:V)mixture significantly improved the extraction efficiency while minimizing matrix effect.A hybridized analytical strategy integrating nontarget and suspect screening was developed to achieve comprehensive identification and classification of pollutants,employing the National Institute of Standards and Technology(NIST)20 library and Agilent Technologies Personal Compound Database and Library(PCDL).This approach successfully characterized 273 organic contaminants spanning 12 categories,including polycyclic aromatic hydrocarbons(PAHs)and their derivatives,esters,and phenolic compounds in human serum,with a significant increase in detection specificity compared to conventional workflows.The methodology used serum samples of the workers from coking industry,revealing widespread contamination dominated by PAHs and PAH derivatives.Among the target analytes,three were identified solely by NIST and six solely by PCDL,indicating the complementary benefits of combining these different databases.Notably,this work reported the first confirmed detection of 2-naphthalenamine in human serum.This optimized approach demonstrates enhanced sensitivity and reliability in serum analysis,advancing biomonitoring capabilities and providing a deep understanding of human exposure to environmental pollutants.

摘要Understanding the impacts of human activities on the plateau’s living environment is essential for advancing modernization pathways that promote harmony between humanity and nature.However,studies on the dynamic interactions between human activities and the living environment on the Qinghai-Xizang Plateau(QXP)remain limited,with a paucity of quantitative relationship analyses.This study established an assessment framework to evaluate human influences on the living environment in QXP,using data on typical human activities,ecological conditions,and human settlements.Within this framework,the spatial analysis methods and the coupling coordination model were used to examine the spatio-temporal characteristics and relationship of human activities and living environment on the QXP from 2000 to 2020.The geographical detector model was then applied to identify the key factors influencing the plateau’s human living environment.Subsequently,the four-quadrant analysis model was adopted to assess human influences on the living environment.The results indicate that the human activity intensity(HAI)on the QXP remained relatively low yet increased by 15.41%from 2000 to 2020.Spatially,the human living environment quality(LEQ)improved from northwest to southeast,with 61.14%of the areas remaining stable and 18.47%experiencing slight improvement.The analysis of coupling coordination revealed a continuous improvement between the HAI and LEQ,with the areas of high and relatively high coordinated types increasing by more than 9%.Precipitation and urban-rural construction were identified as the primary factors influencing changes in the LEQ.The interaction between the HAI and LEQ was strengthening,with 40.44%classified as coordinated development type and 38.35%as development-environment conflict type.These findings provide valuable insights for enhancing the resilience of human settlements and promoting green development across the plateau.

摘要With the growing advancement of wireless communication technologies,WiFi-based human sensing has gained increasing attention as a non-intrusive and device-free solution.Among the available signal types,Channel State Information(CSI)offers fine-grained temporal,frequency,and spatial insights into multipath propagation,making it a crucial data source for human-centric sensing.Recently,the integration of deep learning has significantly improved the robustness and automation of feature extraction from CSI in complex environments.This paper provides a comprehensive review of deep learning-enhanced human sensing based on CSI.We first outline mainstream CSI acquisition tools and their hardware specifications,then provide a detailed discussion of preprocessing methods such as denoising,time–frequency transformation,data segmentation,and augmentation.Subsequently,we categorize deep learning approaches according to sensing tasks—namely detection,localization,and recognition—and highlight representative models across application scenarios.Finally,we examine key challenges including domain generalization,multi-user interference,and limited data availability,and we propose future research directions involving lightweight model deployment,multimodal data fusion,and semantic-level sensing.

摘要Aldehyde dehydrogenase 2(ALDH2),a mitochondrial enzyme,is the main acetaldehyde dehydrogenase involved in the scavenging of alcohol-derived acetaldehyde and endogenous aldehydes.The ALDH2rs671mutation affects 560 million East Asians and is closely related to an increased risk of various human diseases.In addition to its well-known function in detoxifying alcohol-derived acetaldehyde and endogenous aldehydes,ALDH2 is implicated in human health through its regulation of autophagic machinery and multiple cell death pathways(e.g.,apoptosis,necroptosis,pyroptosis,ferroptosis,and NETosis).This review summarizes the current knowledge of ALDH2 and the regulatory mechanism through which ALDH2 regulates autophagy and cell death.In addition,we outline the potential role of ALDH2 in the regulation of autophagy and cell death during the occurrence and progression of human diseases,aiming to provide a novel theoretical framework for human disease treatment.

摘要The rapid progress of human organoid toolkits presents unprecedented opportunities in disease modeling,drug discovery,and personalized therapy,alongside profound bioethical challenges and concerns.On April 29th,2025,China’s National Science and Technology Ethics Committee(Life Science Ethics Subcommittee)issued the Human Organoid Research Ethical Guidelines[1],establishing the world’s first comprehensive governance framework,especially focusing on brain organoids,embryo models,and chimeric research.This Guidelines represent a pioneering governance model in emerging biotechnology.This commentary offers an in-depth analysis to examine the policy’s innovative three-tiered structure,contrast it with current global regulatory standards,evaluate its domestic impacts as well as potential implications for international biotechnology governance,and provide recommendations for future directions.

摘要Human spinal cord organoids(hSCOs)offer a promising platform to study neurotrauma by addressing many limitations of traditional research models.These organoids provide access to human-specific physiological and genetic mechanisms and can be derived from an individual's somatic cells(e.g.,blood or skin).This enables patient-specific paradigms for precision neurotrauma research,pa rticula rly relevant to the over 300,000 people in the United States living with chronic effects of spinal cord injury(SCI).

摘要Neonicotinoid insecticides(NEOs)are a group of widely used insecticides in agriculture,livestock,and pest control for pets.However,increasing reports on their adverse effects on non-target organisms have aroused growing public concerns.Therefore,it is necessary to evaluate NEO exposure and the related human health.This review summarized the human biomonitoring studies on NEOs,and the epidemiological,in vitro,and in vivo toxicological investigations on the health effects of NEOs.Human biomonitoring studies revealed significant changes in NEO detection frequencies,concentrations,and profiles across different human populations,suggesting potential variances in exposure patterns or metabolic processes.Certain metabolites may be more sensitive biomarkers than their parent molecules,as evidenced by higher levels of some NEO metabolites than those of their parent compounds.Although human epidemiological data has revealed associations between NEO exposure and diverse detrimental health outcomes including reproductive and developmental abnormalities,oxidative damage,etc.,the cross-sectional design of most studies cannot elucidate causality,necessitating further prospective research.In vitro and in vivo studies have shown various toxic effects of NEOs,including neurotoxicity,hepatotoxicity,reproductive toxicity,genotoxicity,etc.Further research on the molecular mechanisms beyond oxidative stress is worthwhile.Given metabolic activation,additional metabolites of both conventional and new generation NEOs as well as NEO-like compounds need to be determined,and their toxicities should be evaluated.This review can be contributed to the comprehensive understanding of the threat from extensive exposure to NEOs and proposed future research directions can aid to improve studies on health effects of NEOs.
