BACKGROUND Schizophrenia presents complex challenges in older patients due to cognitive and social decline.Existing drug treatments offer limited benefits and pose risks,while aerobic exercise shows promise as a noninvasive intervention whose impact remains underexplored.AIM To investigate the effects of aerobic exercise on cognitive and social function in older patients with schizophrenia.METHODS A retrospective study was conducted in 158 older patients with schizophrenia treated at The First Affiliated Hospital of Chongqing Medical and Pharmaceutical College(June 2023 and December 2024).The patients were divided into an observation group(n=86),which received 3 months of aerobic rehabilitation alongside routine treatment,and a control group(n=72),which received routine treatment alone.Cognitive function was assessed using the Measurement and Treatment Research to Improve Cognition in Schizophrenia Consensus Cognitive Battery,social function using the Inpatient Psychiatric Rehabilitation Outcome Scale(IPROS),and psychotic symptoms using the Positive and Negative Syndrome Scale.RESULTS Post-treatment,Positive and Negative Syndrome Scale total scores were lower in the observation group than in the controls(P<0.001),with significant improvements in positive,negative,and general symptoms.Measurement and Treatment Research to Improve Cognition in Schizophrenia Consensus Cognitive Battery domain scores were improved in processing speed,working memory,verbal/visual learning,and executive function(all P<0.05).IPROS total scores were decreased(P<0.001),indicating better social functioning.Longer exercise duration correlated with greater cognitive gains and lower social function deficits.Body mass index declined(P<0.001),and adverse events were mild and transient(9.3%).Multivariate linear regression confirmed that aerobic exercise was independently associated with a significant reduction in IPROS scores(Adjustedβ=-4.57,95%confidence intervals:-6.08 to-3.05,P<0.001).CONCLUSION Aerobic exercise effectively improves cognitive function,social function,and psychotic symptoms in older patients with schizophrenia,is safe,and serves as a valuable adjunctive intervention. 展开更多

Graphitic carbon nitride(g-CN)stands out as the most promising candidate for solar energy conversion owing to its easy preparation,metal-free nature,flexible molecular structure,moderate bandgap,and excellent thermal/chemical stability.To enhance the performance of intrinsic g-CN,a supramolecular self-assembly strategy has been proposed to regulate the molecular structure of supramolecular precursors through non-covalent interactions across molecular building blocks,thereby optimizing the electronic structure of g-CN.This review provides a comprehensive overview of the recent progress in supramolecular self-assembly-derived graphitic carbon nitride(SM-CN)from both experimental and theoretical computational research in synthesis strategies,including synthesis methods and influencing factors,providing a theoretical foundation for the design of supramolecular assembly.It also discusses modification strategies,such as internal modification of the conjugated plane,interlayer optimization,and construction of heterointerfaces to improve the electronic structure of SM-CN owing to its unique layered structure.This review further summarizes the applications of SM-CN in environment and energy,including wastewater treatment,sterilization and disinfection/air purification,water splitting,H2O2production,organic synthesis/biomass conversion,CO2reduction,photocatalytic coupling technology.Finally,perspectives and outlooks for the future development of SM-CN aim to inspire further innovation in the design and construction of high-performance SM-CN for broader applications. 展开更多

Chronic obstructive pulmonary disease(COPD),a disease responsible for early mortality worldwide,is well accepted to be associated with periodontitis epidemiologically.Although both of the diseases are the multi-microbial inflammatory disease,the precise underlying mechanisms by which periodontitis influences the progression of COPD remains largely unknown.Here,we established COPD accompanied with periodontitis mouse models and observed the pronounced progress in pulmonary symptoms and histopathology,cha racterized by poorer respiratory function,thicke ned bronchial walls,and increased neutrophils infiltration in lung tissue.Mechanistically,periodontitis pathogen Porphyromonas gingivalis(P.gingivalis)relocated in the lung through the respiratory tract and LPS from P.gingivalis promoted the secretion of chemokines CXCL2 and G-CSF of alveolar epithelial cells through NF-κB and p38 MAPK pathways to recruit neutrophils.Furthermore,exposure to P.gingivalis of infiltrated neutrophils released matrix metallopeptidase-8(MMP-8)and neutrophil elastase(NE),which aggravated airway inflammation and tissue damage.These findings indicated that periodontitis could exacerbate COPD via its pathogen P.gingivalis,which translocated in the lung and stimulated neutrophil chemotaxis and activation in the lung. 展开更多

Functional near-infrared spectroscopy quantifies cerebral hemodynamic signals by capturing oxygenation-dependent changes in hemoglobin in a noninvasive,portable,and ecologically valid manner,providing a unique insight into neurovascular coupling.However,functional imaging biomarkers with high ecological validity for neurological disorders such as stroke,Parkinson's disease,dementia,amyotrophic lateral sclerosis,epilepsy,spinal cord injury,and traumatic brain injury are lacking,limiting the mechanistic understanding,treatment evaluations,and individualized interventions.The aim of this review is to systematically summarize evidence from the past decade on the use of functional near-infrared spectroscopy under the aforementioned conditions,synthesize its value for revealing neural mechanisms and assessing therapeutic responses,and identify current technical bottlenecks and future directions for advancement.Collectively,the findings demonstrate that functional near-infrared spectroscopy possesses substantial and far-reaching potential for uncovering the neural mechanisms underlying disease and for evaluating treatment-induced changes in brain function.Equipped with wearable probes,functional near-infrared spectroscopy can continuously and noninvasively monitor brain activity in naturalistic environments for extended periods,thereby overcoming the limitations of conventional imaging modalities that can only acquire data under restricted settings.This capability can furnish unprecedented objective neuroimaging evidence for neuroregenerative therapy research.Moreover,the portability of functional near-infrared spectroscopy allows it to be integrated into neurofeedback training systems:hemoglobin signals can be fed back to participants within milliseconds,enabling targeted,individualized,closed-loop modulation of brain function and considerably expanding the scope of hemodynamicsbased neurofeedback.When combined with other brain function assays(such as electroencephalography)and intervention techniques(such as transcranial magnetic stimulation and transcranial direct current stimulation),functional near-infrared spectroscopy also supplies high-temporal-resolution hemodynamic information,laying a critical foundation for the construction of high-precision noninvasive brain–computer interfaces,real-time cognitivestate decoding,and adaptive neuromodulation.Admittedly,almost all existing functional near-infrared spectroscopy studies are still observational and have small sample sizes,short follow-ups,and insufficient controls—shortcomings that together produce low-grade evidence.Therefore,there is still a significant gap before clinical translation can be achieved.Technically,the limited penetration depth of functional near-infrared spectroscopy restricts sampling to the superficial cortex,leaving deep nuclei largely unreachable.In addition,no consensus exists across devices regarding optode layout,light-source choice,motion-artifact correction,or analytical pipelines,creating pronounced heterogeneity that undermines reproducibility.With artificial intelligence and big data analytics advancing rapidly,functional near-infrared spectroscopy embedded within multimodal fusion frameworks is now poised to systematically map aberrant brain function signatures of neurological disorders,identify pathological regions suitable for targeted intervention,and provide real-time assessments of functional changes produced by neuroregenerative therapies. 展开更多

A growing global population and the increasing prevalence of diet-related health issues such as“hidden hunger”,obesity,hypertension,and diabetes necessitate a fundamental rethinking of crop design and breeding.Synthetic metabolic engineering offers a method to modify and redesign metabolic pathways to increase the nutritional value of crops.We summarize recent advances in the biofortification of key nutrients including provitamin A,vitamin C,vitamin B9,iron,zinc,anthocyanins,flavonoids,and unsaturated fatty acids.We discuss the potential of multi-gene stacking,gene editing,enzyme engineering,and artificial intelligence in synthetic metabolic engineering.We propose future research directions and potential solutions centered on leveraging AI-driven systems biology,precision gene editing,enzyme engineering,agrobacterium-mediated genotype-independent transformation,and modular metabolic engineering strategies to develop next-generation nutritionally enhanced super crops and transform global food systems. 展开更多

Understanding the properties of warm dense hydrogen is of key importance for the modeling of compact astrophysical objects and to understand and further optimize inertial confinement fusion applications.The workhorse of warm dense matter theory is thermal density functional theory(DFT),which,however,suffers from two limitations:(i)its accuracy can depend on the utilized exchange-correlation functional,which has to be approximated,and(ii)it is generally limited to single-electron properties such as the density distribution.Here,we present a new ansatz combining time-dependent DFT results for the dynamic structure factor See(q,ω)with static DFT results for the density response.This allows us to estimate the electron-electron static structure factor See(q)of warm dense hydrogen with high accuracy over a broad range of densities and temperatures.In addition to its value for the study of warm dense matter,our work opens up new avenues for the future study of electronic correlations exclusively within the framework of DFT for a host of applications. 展开更多

BACKGROUND Cognitive impairment is a common functional impairment after stroke that severely affects the quality of life of patients.The underlying neurobiological mechanisms of poststroke cognitive impairment(PSCI)remain unclear.AIM To investigate the changes in functional connectivity(FC)in the brains of patients with PSCI.METHODS A total of 21 patients with PSCI and 12 healthy controls were selected as study subjects,and resting-state functional magnetic resonance imaging was performed.The brain region[Cerebellum_6_R(aal)]with significant differences identified by regional homogeneity analysis and the left thalamus,right thalamus,left basal ganglia,and right basal ganglia in the Brainnetome Atlas were selected as the seeds(regions of interest),and the FC between the seeds and whole-brain voxels was analyzed.Moreover,the 116 brain regions defined in the AAL116 atlas were selected as seeds(regions of interest),and the FC between the whole-brain seeds was calculated.RESULTS The results of the seed-based FC analysis revealed that the FC of the Cerebelum_9_R,Occipital_Mid_L,and Fusiform_R in the PSCI group was significantly greater than that in the control group.FC analysis of whole-brain seeds revealed that the FC of 20 pairs(Cerebelum_4_5_R and Cerebelum_6_R,etc.)in the PSCI group was significantly greater than that in the healthy control group.CONCLUSION Patients with PSCI exhibit changes in the FC of specific brain regions in the resting state,which may help researchers explore the underlying neurobiological mechanisms of PSCI from a new perspective. 展开更多

Ln@MOFs by anchoring rare metal ions(Ln) into metal-organic frameworks(MOFs) are proved to have great potential in the field of luminescent molecular thermometer.Nevertheless,the current research indicated that the poor structural stability and low sensitivity hindered their application scope.In this work,a new MOF Zn-450 luminescent thermometer with multiple emission fluorescence characteristics was synthesized by the combination of 3,3,5,5-biphenyl tetracarboxylic acid(H4L) and Zn2+ ion under solvothermal conditions.Interestingly,a high relative sensitivity of 1.43 % K-1 was found within 80-300 K based on Zn-450.Subsequently,two high-sensitivity luminescent Ln@MOFs(Ln = Eu and Tb) were further fabricated by doping rare earth ions into Zn-450 based on the post-synthesis strategy.Among them,the Eu@Zn-450 demonstrates various luminous behaviors while achieving an increased relative sensitivity of 1.63 % K-1.In addition,the continuously visible red,pink,and purple luminescent emissions at the same temperature range were observed,suggesting that the Eu@Zn-450 could be utilized as a luminescent colorimetric molecular thermometer.Importantly,this work can present new possibilities for the development of rare earth-doped luminescence and its temperature sensing properties. 展开更多

Background Weaning-induced diarrhoea and growth retardation in piglets are associated with impaired intestinal barrier function and decreased levels of colonic short-chain fatty acids(SCFAs).Although SCFA supplementation has been proposed to mitigate these issues,the efficacy and optimal dosage of sodium isobutyrate remain unclear.Results We investigated the effects of sodium isobutyrate supplementation(500,1,000,2,000,and 4,000 mg/kg diet)on weaned piglets(Duroc×Landrace×Yorkshire,28 d of age;n=8).After a 28-d feeding trial,supplementation at 500–2,000 mg/kg significantly improved average daily gain and feed efficiency and reduced diarrhoea frequency,with maximal benefits observed at 1,000 mg/kg(P<0.0001).Additionally,500–1,000 mg/kg sodium isobutyrate supplementation increased the apparent digestibility of crude protein,organic matter,and crude fibre(P<0.05).Serum biochemical parameters were unaffected,although secretory immunoglobulin A(SIgA)levels significantly increased upon supplementation with 500–1,000 mg/kg(P<0.05).16S rRNA gene sequencing indicated that sodium isobutyrate increased the abundance of beneficial colonic microbiota.The 1,000 mg/kg group presented the most pronounced effect,with a significant increase of the relative abundance of Prevotella and the greatest improvement in SCFA concentrations(P<0.05).Metabolomics revealed elevated levels of colonic indole-3-lactic acid and 3-hydroxybutyrate upon supplementation with 1,000 mg/kg(P<0.05).Transcriptomic analyses indicated activation of protein digestion and absorption pathways,and PI3K-Akt signalling,marked by TSG-6 upregulation and the suppression of ISG15 and DDIT4 expression(P<0.05).Supplementation with 1,000 mg/kg was associated with improved intestinal barrier-related markers,including reduced serum D-lactate,diamine oxidase,and lipopolysaccharide levels,increased tight junction protein expression;activation of G protein-coupled receptors;and inhibition of TLR4/MyD88/NF-κB signalling(P<0.05),suggesting enhanced barrier function.Conclusions In conclusion,dietary supplementation with 1,000 mg/kg sodium isobutyrate was associated with improved intestinal morphology,reduced serum permeability,increased expression of tight junction proteins,and enhanced immune function in weaned piglets,suggesting enhanced colonic barrier function and providing dosage guidance and mechanistic insights for future applications. 展开更多

Background:Rats are often used to prepare skin defect models.However,the skin defect sizes of the models prepared by researchers are different,and the lack of consensus on the critical-size defect makes it difficult to compare their research results.Methods:The time for wound closure was evaluated and recorded through gross observation.The regression equation between the healing time and the diameter of skin defect was established,which can be used to predict the healing time for a certain skin defect size in rats.Histochemical and immunohistochemical staining was used to observe the regeneration and reconstruction of skin appendages,and the functional skin repair was quantitatively scored.Results:The critical-size defect of rats was determined based on the maximum capacity of structural skin repair,and the functional skin repair was quantitatively scored based on the regeneration and reconstruction of skin appendages.The allowable range of critical-size skin defect of SD rats lies between 45 and 50 mm in diameter.The concept of structural repair and the category of functional repair of injured skin are put forward.The regression equation between the structural skin healing time and defect diameters is established.Conclusion:The allowable range of skin critical-size defect of SD rats lies between 45 and 50 mm in diameter.The regression equation between the structural skin healing time and defect diameters can be used to predict the healing time for a certain skin defect size in rats. 展开更多

Current seismic damage assessments for high-speed railway(HSR)bridges primarily focus on the overall structural safety,lacking evaluations from multiple performance perspectives,which affects the post-earthquake traffic decision-making for the bridges.This study proposes a performance-based comprehensive functional damage probability assessment framework for high-speed railway simply supported bridges(HSRSSBs)under earthquakes.The framework categorizes the functions of HSR bridges into three levels:post-earthquake traffic function(PTF),structural bearing function(SBF),and collapse resistance function(CRF),corresponding to the operational,structural safety,and structural integrity requirements of HSRSSB,respectively.By analyzing the damage states of key bridge components during earthquakes,the functional damage probability assessment indicators and classification thresholds are established according to various performance requirements.Damage probability calculations are conducted using the probability density evolution method and vulnerability method.Finally,based on the relationship between damage probabilities at different functional levels,a comprehensive damage probability assessment framework considering the three-level performance requirements of HSRSSBs is developed,and the influence of varying pier heights on the functional damage probability relationship is examined.The results indicate that current HSRSSB designs meet all performance requirements under frequent earthquakes.Under design-level earthquake conditions,the SBF remains in a slight damage state,while the PTF exhibits varying degrees of damage,which worsens as pier height increases.The pier structure satisfies seismic demands even under rare earthquake conditions. 展开更多

Spinal cord injury represents a severe form of central nervous system trauma for which effective treatments remain limited.Microglia is the resident immune cells of the central nervous system,play a critical role in spinal cord injury.Previous studies have shown that microglia can promote neuronal survival by phagocytosing dead cells and debris and by releasing neuroprotective and anti-inflammatory factors.However,excessive activation of microglia can lead to persistent inflammation and contribute to the formation of glial scars,which hinder axonal regeneration.Despite this,the precise role and mechanisms of microglia during the acute phase of spinal cord injury remain controversial and poorly understood.To elucidate the role of microglia in spinal cord injury,we employed the colony-stimulating factor 1 receptor inhibitor PLX5622 to deplete microglia.We observed that sustained depletion of microglia resulted in an expansion of the lesion area,downregulation of brain-derived neurotrophic factor,and impaired functional recovery after spinal cord injury.Next,we generated a transgenic mouse line with conditional overexpression of brain-derived neurotrophic factor specifically in microglia.We found that brain-derived neurotrophic factor overexpression in microglia increased angiogenesis and blood flow following spinal cord injury and facilitated the recovery of hindlimb motor function.Additionally,brain-derived neurotrophic factor overexpression in microglia reduced inflammation and neuronal apoptosis during the acute phase of spinal cord injury.Furthermore,through using specific transgenic mouse lines,TMEM119,and the colony-stimulating factor 1 receptor inhibitor PLX73086,we demonstrated that the neuroprotective effects were predominantly due to brain-derived neurotrophic factor overexpression in microglia rather than macrophages.In conclusion,our findings suggest the critical role of microglia in the formation of protective glial scars.Depleting microglia is detrimental to recovery of spinal cord injury,whereas targeting brain-derived neurotrophic factor overexpression in microglia represents a promising and novel therapeutic strategy to enhance motor function recovery in patients with spinal cord injury. 展开更多

Diabetic retinopathy is a prominent cause of blindness in adults,with early retinal ganglion cell loss contributing to visual dysfunction or blindness.In the brain,defects inγ-aminobutyric acid synaptic transmission are associated with pathophysiological and neurodegenerative disorders,whereas glucagon-like peptide-1 has demonstrated neuroprotective effects.However,it is not yet clear whether diabetes causes alterations in inhibitory input to retinal ganglion cells and whether and how glucagon-like peptide-1 protects against neurodegeneration in the diabetic retina through regulating inhibitory synaptic transmission to retinal ganglion cells.In the present study,we used the patch-clamp technique to recordγ-aminobutyric acid subtype A receptor-mediated miniature inhibitory postsynaptic currents in retinal ganglion cells from streptozotocin-induced diabetes model rats.We found that early diabetes(4 weeks of hyperglycemia)decreased the frequency of GABAergic miniature inhibitory postsynaptic currents in retinal ganglion cells without altering their amplitude,suggesting a reduction in the spontaneous release ofγ-aminobutyric acid to retinal ganglion cells.Topical administration of glucagon-like peptide-1 eyedrops over a period of 2 weeks effectively countered the hyperglycemia-induced downregulation of GABAergic mIPSC frequency,subsequently enhancing the survival of retinal ganglion cells.Concurrently,the protective effects of glucagon-like peptide-1 on retinal ganglion cells in diabetic rats were eliminated by topical administration of exendin-9-39,a specific glucagon-like peptide-1 receptor antagonist,or SR95531,a specific antagonist of theγ-aminobutyric acid subtype A receptor.Furthermore,extracellular perfusion of glucagon-like peptide-1 was found to elevate the frequencies of GABAergic miniature inhibitory postsynaptic currents in both ON-and OFF-type retinal ganglion cells.This elevation was shown to be mediated by activation of the phosphatidylinositol-phospholipase C/inositol 1,4,5-trisphosphate receptor/Ca2+/protein kinase C signaling pathway downstream of glucagon-like peptide-1 receptor activation.Moreover,multielectrode array recordings revealed that glucagon-like peptide-1 functionally augmented the photoresponses of ON-type retinal ganglion cells.Optomotor response tests demonstrated that diabetic rats exhibited reductions in visual acuity and contrast sensitivity that were significantly ameliorated by topical administration of glucagon-like peptide-1.These results suggest that glucagon-like peptide-1 facilitates the release ofγ-aminobutyric acid onto retinal ganglion cells through the activation of glucagon-like peptide-1 receptor,leading to the de-excitation of retinal ganglion cell circuits and the inhibition of excitotoxic processes associated with diabetic retinopathy.Collectively,our findings indicate that theγ-aminobutyric acid system has potential as a therapeutic target for mitigating early-stage diabetic retinopathy.Furthermore,the topical administration of glucagon-like peptide-1 eyedrops represents a non-invasive and effective treatment approach for managing early-stage diabetic retinopathy. 展开更多

Turnout irregularity significantly affects the stochastic vibration behavior of vehicle-turnout structures.This study proposes a fitting formula for the turnout irregularity spectrum and develops a turnout irregularity full information expression model(TIFIEM)using a stochastic harmonic function.The model is applied to vehicle-turnout structure stochastic vibration and reliability analysis.Findings suggest that the Hamming window method,with a window length of 4096 points,is optimal for estimating the turnout irregularity spectrum.It is recommended to fit the power spectral density(PSD)using a 5th-order polynomial for better accuracy.The TIFIEM effectively addresses randomness in amplitude,frequency,and phase.An analysis of 250 irregularity samples is sufficient for the desired accuracy.Additionally,the PSD amplitude at various frequency points follows a Chi-square distribution with 2°of freedom.Regions 3-7 m from the tip of the switch rail on the straight switch rail and 53-54 m on the point rail are most susceptible to wear.When the vehicle passes through the turnout at 300 km/h,the reliability of vehicle-turnout structures at the crossing panel decreases to 95.8%. 展开更多

Spontaneous recovery frequently proves maladaptive or insufficient because the plasticity of the injured adult mammalian central nervous system is limited.This limited plasticity serves as a primary barrier to functional recovery after brain injury.Neuromodulation technologies represent one of the fastest-growing fields in medicine.These techniques utilize electricity,magnetism,sound,and light to restore or optimize brain functions by promoting reorganization or long-term changes that support functional recovery in patients with brain injury.Therefore,this review aims to provide a comprehensive overview of the effects and underlying mechanisms of neuromodulation technologies in supporting motor function recovery after brain injury.Many of these technologies are widely used in clinical practice and show significant improvements in motor function across various types of brain injury.However,studies report negative findings,potentially due to variations in stimulation protocols,differences in observation periods,and the severity of functional impairments among participants across different clinical trials.Additionally,we observed that different neuromodulation techniques share remarkably similar mechanisms,including promoting neuroplasticity,enhancing neurotrophic factor release,improving cerebral blood flow,suppressing neuroinflammation,and providing neuroprotection.Finally,considering the advantages and disadvantages of various neuromodulation techniques,we propose that future development should focus on closed-loop neural circuit stimulation,personalized treatment,interdisciplinary collaboration,and precision stimulation. 展开更多

Freezing of gait is a significant and debilitating motor symptom often observed in individuals with Parkinson's disease.Resting-state functional magnetic resonance imaging,along with its multi-level feature indices,has provided a fresh perspective and valuable insight into the study of freezing of gait in Parkinson's disease.It has been revealed that Parkinson's disease is accompanied by widespread irregularities in inherent brain network activity.However,the effective integration of the multi-level indices of resting-state functional magnetic resonance imaging into clinical settings for the diagnosis of freezing of gait in Parkinson's disease remains a challenge.Although previous studies have demonstrated that radiomics can extract optimal features as biomarkers to identify or predict diseases,a knowledge gap still exists in the field of freezing of gait in Parkinson's disease.This cross-sectional study aimed to evaluate the ability of radiomics features based on multi-level indices of resting-state functional magnetic resonance imaging,along with clinical features,to distinguish between Parkinson's disease patients with and without freezing of gait.We recruited 28 patients with Parkinson's disease who had freezing of gait(15 men and 13 women,average age 63 years)and 30 patients with Parkinson's disease who had no freezing of gait(16 men and 14 women,average age 64 years).Magnetic resonance imaging scans were obtained using a 3.0T scanner to extract the mean amplitude of low-frequency fluctuations,mean regional homogeneity,and degree centrality.Neurological and clinical characteristics were also evaluated.We used the least absolute shrinkage and selection operator algorithm to extract features and established feedforward neural network models based solely on resting-state functional magnetic resonance imaging indicators.We then performed predictive analysis of three distinct groups based on resting-state functional magnetic resonance imaging indicators indicators combined with clinical features.Subsequently,we conducted 100 additional five-fold cross-validations to determine the most effective model for each classification task and evaluated the performance of the model using the area under the receiver operating characteristic curve.The results showed that when differentiating patients with Parkinson's disease who had freezing of gait from those who did not have freezing of gait,or from healthy controls,the models using only the mean regional homogeneity values achieved the highest area under the receiver operating characteristic curve values of 0.750(with an accuracy of 70.9%)and 0.759(with an accuracy of 65.3%),respectively.When classifying patients with Parkinson's disease who had freezing of gait from those who had no freezing of gait,the model using the mean amplitude of low-frequency fluctuation values combined with two clinical features achieved the highest area under the receiver operating characteristic curve of 0.847(with an accuracy of 74.3%).The most significant features for patients with Parkinson's disease who had freezing of gait were amplitude of low-frequency fluctuation alterations in the left parahippocampal gyrus and two clinical characteristics:Montreal Cognitive Assessment and Hamilton Depression Scale scores.Our findings suggest that radiomics features derived from resting-state functional magnetic resonance imaging indices and clinical information can serve as valuable indices for the identification of freezing of gait in Parkinson's disease. 展开更多

Objectives:Mitochondrial function is intricately linked to osteogenic and osteoclastic differentiation.The mitochondrial calcium uniporter(MCU)is a critical regulator of mitochondrial function,influencing key aspects of cellular metabolism and signaling.However,the precise mechanisms by which MCU modulates osteogenic activity remain unclear.This study aimed to elucidate the impact of MCU-mediated regulation of mitochondrial function on bone remodeling and to explore the underlying mechanisms.Methods:The mouse pre-osteoblastic cells(MC3T3-E1)were treated with the MCU-specific inhibitor Ru265 during osteogenic induction to assess changes in osteogenic differentiation capacity,mitochondrial function,and mitochondrial dynamics.Additionally,MCU global knockout(MCU KO)mice were employed as an in vivo model to explore the role of MCU in bone structure phenotype through bone microstructural analysis and histological examination.Results:Quantitative reverse transcription(qRT)PCR,western blotting,alizarin Red-S(ARS)staining,and alkaline phosphatase(ALP)activity analyses revealed that the inhibition of MCU function by Ru265 downregulates ALP activity(about 59.60%of the control group)and the expression of osteogenic markers in MC3T3-E1 cells.Dramatically increased dynamin-related protein 1(Drp1)expression(about 1.13 times of the control group),decreased mitofusion-2(Mfn2)expression(about 14.51%of the control group),and reduced mitochondrial membrane potential(MMP)(about 55.16%of the control group)were observed,all indicating substantial disruption of mitochondrial dynamics and function in MC3T3-E1 cells.The corroborating evidence is thatμCT and histological analyses of MCU global knockout mice revealed impaired osteogenic differentiation,reduced bone mass formation,and deteriorated trabecular bone microstructure compared with wild-type mice.Conclusion:MCU inhibition elicits aberrant mitochondrial dynamics and mitochondrial dysfunction,thereby impairing osteogenic function and disrupting bone remodeling,which could have promising implications for bone metabolism. 展开更多

Iron(Fe)and manganese(Mn)mixed oxide composites are widely recognized for their excellent adsorption performance in wastewater treatment.However,their functionalization for simultaneous removal of both anionic and cationic contaminants remains underexplored.In this study,Fe-Mn mixed oxides were functionalized with poly(acrylic acid)(PAA)and chitosan to introduce carboxylic and amine groups respectively,forming a dual-functionalized composite(PAA-chitosan-FeMn).The material was evaluated for the adsorption of Congo red,reactive blue 19,and methylene blue from aqueous solutions using batch and fixed-bed reactor systems.The composite was characterized before and after adsorption using X-ray photoelectron spectroscope(XPS),atomic force microscope,scanning electron microscope,high-resolution transmission electron microscope,Fourier transform infrared spectroscope(FTIR),thermal gravimetric analyzer,and Raman spectroscopy.A marked enhancement in adsorption capacity was observed for all target dyes.Mechanistic investigations using XPS,FTIR,and density functional theory(DFT)indicated that the adsorption process was predominantly governed by electrostatic interactions and hydrogen bonding involving carbon,nitrogen,and oxygen functional groups.XPS analysis further confirmedthe active role of Fe and Mn ions in dye binding.Comprehensive studies on adsorption isotherms,kinetics,and thermodynamics were conducted under optimized conditions.The composite exhibited a specificsurface area of 27.4 to 68.4 m2·g-1before and after dye adsorption and a point of zero charge of 6.5-6.8,facilitating effective adsorption of oppositely charged dyes at nearneutral pH.Batch adsorption experiments demonstrated maximum capacities of 95.0%to 99.7% for both dyes under optimal conditions(pH 8-10,298 K).The PAA-chitosan-FeMn composite demonstrated high adsorption capacity,excellent stability under acidic conditions,and strong selectivity,highlighting its potential for the effective and simultaneous removal of anionic and cationic azo dyes from wastewater. 展开更多

BACKGROUND Psychosocial factors play a major role in the pathogenesis of functional dyspepsia(FD);however,there is a paucity of evidence from randomized controlled trials supporting the use of neuromodulators in FD.AIM To evaluate the efficacy and safety of lansoprazole(LPZ)combined with flupentixol-melitracen(FM)in FD.METHODS This was a single-center,double-blind,randomized,placebo-controlled trial enrolling patients aged 18-80 years who met the Rome IV diagnostic criteria for FD.Patients with a positive Helicobacter pylori test or abnormal upper gastrointestinal endoscopy/abdominal ultrasound were excluded.Participants completed the Leuven Postprandial Distress Scale,Short Form Nepean Dyspepsia Index(SFNDI),Patient Health Questionnaire-9(PHQ-9),Generalized Anxiety Disorders-7,and Pittsburgh Sleep Quality Index.Patients were randomized 1:1 to receive LPZ(30 mg once daily)combined with FM(flupentixol 0.5 mg+melitracen 10 mg)or matched placebo for 2 weeks,followed by a 4-week follow-up.The primary endpoint was clinical response rate in the intention-to-treat population,defined as a reduction of at least 0.7 in the postprandial discomfort syndrome(PDS)score at the end of treatment among patients with a baseline PDS score of≥1.RESULTS Between March 5 and August 10,2025,183 patients were randomized to receive LPZ+FM(n=92)or LPZ+placebo(n=91).At week 2,the clinical response rate was higher in the LPZ+FM group than in the placebo group(63.0%vs 46.2%;relative risk=1.42,95%confidence interval:1.04-1.93).LPZ+FM resulted in greater reductions in PDS,SF-NDI,PHQ-9,Generalized Anxiety Disorders-7,and Pittsburgh Sleep Quality Index scores.Improvements in PDS,SF-NDI,and PHQ-9 persisted through follow-up.Adverse events were mild and occurred in 32.6%of patients receiving LPZ+FM and 23.1%receiving placebo,with no serious adverse events reported.CONCLUSION Short-term,low-dose FM combined with LPZ is an effective and safe treatment for FD. 展开更多
