The characterization of drug-target interactions is a key component of drug discovery,testing,and development.Affinity chromatography is one approach that can be used for this type of analysis.For instance,this may be done by using an immobilized target as a stationary phase and a drug as the applied solute.This review will discuss the various ways in which affinity chromatographic methods have been used to examine drug-target interactions,with an emphasis on high-performance methods.The general principles of this approach and factors to consider in its use for drug-target interaction analysis will first be examined.Methods based on zonal elution or frontal analysis for binding and competition studies will then be discussed.Various techniques for kinetic studies will next be considered,along with approaches that employ secondary binding agents and hybrid techniques.In each case,the general principles and theory of an approach will be given along with examples of its use in drug-target interaction studies.Advantages or limitations of each approach will be provided as well.This information should make it possible in the future to extend these techniques to other drug-target systems of interest in biomedical research and drug testing or development. 展开更多

The sulfation and decomposition process has proven effective in selectively extracting lithium from lepidolite.It is essential to clarify the thermochemical behavior and kinetic parameters of decomposition reactions.Accordingly,comprehensive kinetic study by employing thermalgravimetric analysis at various heating rates was presented in this paper.Two main weight loss regions were observed during heating.The initial region corresponded to the dehydration of crystal water,whereas the subsequent region with overlapping peaks involved complex decomposition reactions.The overlapping peaks were separated into two individual reaction peaks and the activation energy of each peak was calculated using isoconversional kinetics methods.The activation energy of peak 1 exhibited a continual increase as the reaction conversion progressed,while that of peak 2 steadily decreased.The optimal kinetic models,identified as belonging to the random nucleation and subsequent growth category,provided valuable insights into the mechanism of the decomposition reactions.Furthermore,the adjustment factor was introduced to reconstruct the kinetic mechanism models,and the reconstructed models described the kinetic mechanism model more accurately for the decomposition reactions.This study enhanced the understanding of the thermochemical behavior and kinetic parameters of the lepidolite sulfation product decomposition reactions,further providing theoretical basis for promoting the selective extraction of lithium. 展开更多

Rock slope instability is a prevalent geological hazard that imposes significant adverse impacts on engineering activities.Although existing studies have focused on homogeneous rock slopes,the theoretical models for quantifying the stability of softhard interbedded anti-inclined slopes remain underdeveloped,primarily due to the complex force transfer mechanisms involved.This study proposed a novel theoretical model for the stability analysis of soft-hard interbedded anti-inclined slopes under rainfall conditions.The framework models stratified rock layers as layered cantilever beams with material heterogeneity.Based on the principle of deformation compatibility,it comprehensively accounted for interlayer force transfer and strength degradation resulting from differential deformations among rock layers.Furthermore,it integrated the critical instability length induced by the self-weight of rock layers to determine the fracture depth.The proposed method was validated against engineering case studies and physical model tests,with error falling within an acceptable range.Compared to existing theoretical methods,the proposed method provided a more realistic representation of the slope's stress field.The analysis results demonstrate that rainfall not only reduces the inclination angle of the failure surface but also leads to an approximate 30%decrease in the safety factor.The proposed theoretical model is particularly useful for quickly calculating the stability of soft-hard interbedded anti-inclined rock slope under rainfall conditions,compared to complex and time-consuming numerical simulation calculations. 展开更多

This paper investigates the reliability of internal marine combustion engines using an integrated approach that combines Fault Tree Analysis(FTA)and Bayesian Networks(BN).FTA provides a structured,top-down method for identifying critical failure modes and their root causes,while BN introduces flexibility in probabilistic reasoning,enabling dynamic updates based on new evidence.This dual methodology overcomes the limitations of static FTA models,offering a comprehensive framework for system reliability analysis.Critical failures,including External Leakage(ELU),Failure to Start(FTS),and Overheating(OHE),were identified as key risks.By incorporating redundancy into high-risk components such as pumps and batteries,the likelihood of these failures was significantly reduced.For instance,redundant pumps reduced the probability of ELU by 31.88%,while additional batteries decreased the occurrence of FTS by 36.45%.The results underscore the practical benefits of combining FTA and BN for enhancing system reliability,particularly in maritime applications where operational safety and efficiency are critical.This research provides valuable insights for maintenance planning and highlights the importance of redundancy in critical systems,especially as the industry transitions toward more autonomous vessels. 展开更多

Border-associated macrophages are located at the interface between the brain and the periphery, including the perivascular spaces, choroid plexus, and meninges. Until recently, the functions of border-associated macrophages have been poorly understood and largely overlooked. However, a recent study reported that border-associated macrophages participate in stroke-induced inflammation, although many details and the underlying mechanisms remain unclear. In this study, we performed a comprehensive single-cell analysis of mouse border-associated macrophages using sequencing data obtained from the Gene Expression Omnibus(GEO) database(GSE174574 and GSE225948). Differentially expressed genes were identified, and enrichment analysis was performed to identify the transcription profile of border-associated macrophages. CellChat analysis was conducted to determine the cell communication network of border-associated macrophages. Transcription factors were predicted using the ‘pySCENIC' tool. We found that, in response to hypoxia, borderassociated macrophages underwent dynamic transcriptional changes and participated in the regulation of inflammatory-related pathways. Notably, the tumor necrosis factor pathway was activated by border-associated macrophages following ischemic stroke. The pySCENIC analysis indicated that the activity of signal transducer and activator of transcription 3(Stat3) was obviously upregulated in stroke, suggesting that Stat3 inhibition may be a promising strategy for treating border-associated macrophages-induced neuroinflammation. Finally, we constructed an animal model to investigate the effects of border-associated macrophages depletion following a stroke. Treatment with liposomes containing clodronate significantly reduced infarct volume in the animals and improved neurological scores compared with untreated animals. Taken together, our results demonstrate comprehensive changes in border-associated macrophages following a stroke, providing a theoretical basis for targeting border-associated macrophages-induced neuroinflammation in stroke treatment. 展开更多

Ship emissions significantly impact coastal air quality,with the Yangtze River Delta(YRD)region accounting for approximately 50%of China's total shipping emissions.Using the Community Multiscale Air Quality(CMAQ)model and process analysis(PA)tool,we quantitatively assessed how atmospheric processes(emissions,chemical reactions,transport,and deposition)contribute to PM2.5and O3,and the chemical pathways of O3formation due to ship emissions.Ship emissions significantly enhanced PM2.5concentrations(>5μg/m3)in the coastal areas of Jiangsu province and offshore regions,with diminishing inland effects.Ship-induced NO3-showed greater inland penetration compared to SO42-,which remained near the coast.In coastal cities,aerosol processes,rather than primary emissions,dominated PM2.5formation,highlighting the importance of secondary formation.O3responses varied spatially,showing coastal titration zones but inland enhancements.Process analysis revealed that vertical transport dominated O3distribution in coastal regions(5-10 ppb),whereas chemical processes showed strong negative contributions(below-10 ppb)along shipping routes.The impact exhibited pronounced diurnal variations,peaking during afternoon hours(14:00-17:00 local standard time(LST),up to 10 ppb/h)with morning titration effects(up to-7.5 ppb/h at 08:00 LST).The vertical profile analysis of shipping-related O3showed surface-level O3titration from ship NOxemissions,with impacts extending to higher layers through vertical mixing.Integrated reaction rate analysis revealed that effective O3control in shipping-influenced regions requires coordinated reduction of both NOxand VOCs.These findings provide insights for developing targeted control strategies,particularly for addressing the complex NOx-O3chemistry and secondary PM2.5formation. 展开更多

Dewatering in multilayered aquifers is closely related to stratigraphic configuration.However,previous studies concentrate on the excavation response to dewatering in particular strata,lacking systematic study on the influence of stratum configuration.This study developed a hydro-mechanical coupled numerical model based on the practical engineering of Shanghai Tower deep excavation.The model and input parameters were validated by field measurements.Besides,a parametric study was conducted to investigate the effect of the thickness,permeability and depth of the aquitard on excavation performances.The analysis indicated that settlements in the excavation were insensitive to the thicknesses and permeability of aquitards,while ground surface settlement increased significantly with the increase of the confined aquifer thickness.Based on the numerical analysis,a fit relationship was established to predict the maximum ground settlements of deep excavation in Shanghai considering the excavation depth,groundwater drawdown and distribution of multilayered aquifers.The applicability of the fit equation was verified by field measurements. 展开更多

The hull structure may collapse or deform severely under fire conditions.In this study,the safety of a ship's cabin structure under fire is evaluated using a dual-zone large eddy fire scenario simulation method and a sequential thermo-mechanical coupling analysis method.Taking a three-compartment section of a naval surface ship as a case study,a machinery room fire scenario was simulated and the fire temperature field was analyzed.Through a dedicated data interface,the full-field time-varying temperature loads were mapped to the finite element model of the compartment section,thereby achieving thermo-mechanical coupled analysis of the cabin structure.The effects of thermal expansion on the hull structure under rising fire temperatures were considered in the evaluation of the residual load-bearing capacity of the cabin.The results indicate that the residual load-bearing capacity of the compartment is closely linked to the fire development stage.Temperature not only significantly affects the mechanical properties of steel but also influences the structural load-bearing capacity through thermally stresses. 展开更多

The Wufeng–Longmaxi Formation derives its name from the Upper Ordovician Wufeng Formation and the Lower Silurian Longmaxi Formation,found in sequence in the Sichuan Basin.This formation hosts rich shale gas reservoirs,and its shale gas enrichment patterns are examined in this study using data from 1197 shale samples collected from 14 wells.Five basic and three key parameters,eight in all,are assessed for each sample.The five basic parameters include burial depth and the contents of four mineral types—quartz,clay,carbonate,and other minerals;the three key parameters,representing shale gas enrichment,are total organic carbon(TOC)content,porosity,and gas content.The SHapley Additive exPlanations(SHAP)analysis originated in game theory is used here in an interpretable machine learning framework,to address issues of heterogeneous data structure,noisy relationships,and multi-objective optimization.An evaluation of the ranking,contribution values,and conditions of changes for these parameters offers new quantitative insights into shale gas enrichment patterns.A quantitative analysis of the relationship between data-sets identifies the primary factors controlling TOC,porosity,and gas content of shale gas reservoirs.The results show that TOC and porosity jointly influence gas content;mineral content has a significant impact on both,TOC and porosity;and the burial depth governs porosity which,in turn,affects the conditions under which shale gas is preserved.Input parameter thresholds are also determined and provide a basis for the establishment of quantitative criteria to evaluate shale gas enrichment.The predictive accuracy of the model used in this study is significantly improved by the step-wise addition of two input parameters,namely TOC and porosity,separately and together.Thus,the game theory method in big data-driven analysis uses a combination of TOC and porosity to evaluate the gas content with encouraging results—suggesting that these are the key parameters that indicate source rock and reservoir properties. 展开更多

This study aims to establish an integrated sensitivity analysis framework for optimization and design of the dynamic performance of mechanical systems such as tracked vehicles,by combining the direct differentiation method(DDM)with the linear multibody system transfer matrix method(linear MSTMM).The rigid-flexible coupled multibody system dynamics model of a tracked vehicle is established using the linear MSTMM and validated through the modal test.Building upon the existing DDM-based eigenvalue sensitivity analysis method within the linear MSTMM,the DDM is embedded into it to enable programmable and efficient computation of dynamic response sensitivities for mechanical systems.The proposed approach is used to quantitatively evaluate the sensitivities of both natural vibration characteristics(e.g.,natural frequencies and mode shapes)and transient dynamic responses of the tracked vehicle with respect to system parameters,successfully identifying critical structural parameters.Compared to conventional finite difference methods,the developed methodology eliminates sensitivity to perturbation step sizes.The contributions of this work lie in establishing a unified theoretical foundation and analysis framework for guiding dynamics optimization and design of mechanical systems,and extending the applicability of the linear MSTMM to sensitivity analysis of transient dynamic responses. 展开更多

Red-fleshed fruits are valued for their vibrant color and high anthocyanin content.Pre-harvest fruit bagging enhances fruit peel pigmentation,but its effect on flesh coloration remains poorly characterized.This study revealed that removing bags from‘Gengcunyangtao’red-fleshed peach fruits triggers the rapid and uniform accumulation of anthocyanins in the flesh,resulting in anthocyanin levels that exceed those in unbagged fruits.The exposure to light after bag removal triggered significant increases in anthocyanin levels within 24 h.This was accompanied by the rapid upregulation of light-responsive and flavonoid biosynthetic gene expression levels within 6 h.A metabolomic analysis indicated that anthocyanin precursors,especially p-coumaric acid,accumulated before bag removal,thereby increasing substrate availability for rapid anthocyanin synthesis.On the basis of a weighted gene co-expression network analysis,MYB transcription factors,anthocyanin transporters,glutathione S-transferase,and multidrug and toxic compound extrusion(MATE)were identified as key regulators that coordinate precursor storage along with light-induced transcriptional activation.Notably,PpMYB4 binds to the promoter of PpGSTF14 and activates its expression,thereby promoting anthocyanin accumulation.The study findings elucidated the temporal coordination of metabolic priming and light-responsive transcriptional regulation driving rapid anthocyanin biosynthesis,with possible implications for improving peach fruit flesh coloration. 展开更多

Crumb rubber concrete(CRC)has emerged as a sustainable solution to the environmental challenges posed by rubber waste.This study introduces an advanced mixed-random-aggregate mesoscale model for CRC based on the Base Force Element Method(BFEM)and the complementary energy principle.The model incorporates different rubber substitution ratios(0%-30%),rubber particle sizes(2 mm and 4 mm),and specimen dimensions(edge lengths of 100,150,and 300 mm).These parameters are considered to investigate their effects on the mechanical properties and failure mechanisms of CRC.Accordingly,the numerical results include stress-strain responses,elastic modulus,and damage progression patterns.The results indicate that increasing rubber content leads to a pronounced reduction in both tensile and compressive strengths.This drop occurs mainly because rubber particles have a much lower elastic modulus.Thus,they represent stress concentrators and weak points,which accelerate the sample failure.Moreover,for the same replacement ratio,CRC with smaller rubber particles(2 mm)exhibits lower strength than CRC with larger particles(4 mm).This reduction occurs because the finer particles create more interfacial areas,introducing more sites for defect initiation and micro-cracks.In addition,increasing specimen size is associated with reduced strength and elastic modulus,highlighting size-dependent phenomena in CRC.The numerical predictions show good agreement with experimental results reported in the literature. 展开更多

Sentiment analysis(SA)has evolved from a niche text-classification task into a central problem in natural language processing,spanning multiple domains,modalities,and languages.This survey provides a comprehensive review of sentiment analysis methods from their origins in lexicon-based approaches through classical machine learning,deep learning architectures,pre-trained transformers,and the current era of large language models(LLMs).We formalize the SA problem across multiple granularity levels(document,sentence,and aspect)and present a taxonomy that encompasses classification,regression,aspect-based sentiment analysis(ABSA),emotion detection,and stance detection tasks across diverse domains including movie reviews,product reviews,healthcare,finance,and social media.We review benchmark datasets spanning text-only corpora(IMDb,SST,SemEval series),multimodal benchmarks(CMU-MOSI,CMU-MOSEI,MELD),and domain-specific evaluation suites such as SentiEval.The methodological evolution is traced from VADER and SentiWordNet,through SVM and Naïve Bayes classifiers,CNN and LSTM architectures,BERT and its variants,to modern LLMs including GPT-4,Llama 3,and ModernBERT,with technical details of key architectures and their mathematical formulations.We provide dedicated analyses of chain-ofthought reasoning for implicit sentiment,multimodal fusion strategies,cross-lingual transfer methods,sarcasm and irony detection,explainability through SHAP and LIME,and the emerging challenge of AI-generated fake reviews.A comparative analysis across paradigms reveals that while LLMs achieve strong zero-shot performance,fine-tuned smaller models remain competitive on standard benchmarks,a finding with significant implications for deployment efficiency.We identify persistent open challenges including domain drift,cultural bias,and the model variability problem,and outline future research directions encompassing reasoning-augmented SA,agentic workflows,federated learning,and real-time edge deployment.With coverage of over 130 references spanning two decades of research and 29 new references from 2024 and 2025,this survey provides a unified roadmap for both newcomers and researchers at the frontier of sentiment analysis. 展开更多

The 13th-century Changzhou Mummy,from the Lower Yangtze region in China,is the earliest known East Asian case of an artificially mummified body employing mercury and cinnabar enema without evisceration.This study conducts multidisciplinary research,integrating paleo-radiological,paleo-pathological,paleo-genetic,and paleo-nutritional analysis to investigate the phenotype,genotype,individual life history,and the process of deliberate mummification performed on this individual.We generate a whole genome with 12.7×coverage,revealing potential genetic predisposition for several atherosclerotic cardiovascular diseases(ASCVD).Stable C and N isotope analysis of bones,teeth,and hairs indicates high animal protein consumption as well as terminal illness.Hereditary and dietary risk factors are consistent with the diagnosis of atherosclerosis determined via postmortem examination.Our study,leveraging high-quality ancient DNA,provides a unique opportunity to challenge and rethink the widely accepted consensus on the relationship between atherosclerosis and post-industrial age lifestyles,uncovering unrecognized genetic polymorphisms of ASCVD among ancient individuals,and improving our understanding of the role of genetic factors in the development and evolution of ASCVD. 展开更多

The Palu-Koro Fault is one of the most active left-lateral strike-slip faults in Indonesia.This study integrates gravity data(GGMPlus),GNSS measurements,and seismic analysis to better understand the fault structure and tectonic behavior.The results show that the high First Horizontal Derivative(FHD)value along the fault corresponds to a sharp density gradient related to high tectonic activities.The secondary fault branches and lineaments can also be identified from Tilt Derivative(TDR),emphasizing the fault system's complexity.GNSS data from the INA-CORS network reveals patterns with notable extensional dilatation rates exceeding 100 nanostrain/year in pull-apart basins and compressional zones displaying significant uplift potential.Shear strain measurements highlight regions with deformation rates exceeding 250 nanostrain/year,particularly in the central Palu-Koro Fault segment.Seismicity clustering analysis indicates shallow to intermediate-depth earthquakes along the fault,majority at depths 20-30 km,reflecting lateral tectonic forces align with strike-slip Palu-Koro Fault activities.The integration of gravity,GNSS,and seismicity results critical insights into the fault's heterogeneity,deformation processes,and potential for large seismic events,contributing to enhanced hazard risk assessment and mitigation strategies in this geologically dynamic region. 展开更多

Objective:To systematically analyze the research hotspots and developmental trajectory of acupuncture ophthalmology using bibliometric methods,with the aim of clarifying research trends,collaborative networks,the evolution of thematic focuses,and predicting future directions.Methods:Literature on acupuncture ophthalmology published between January 1,1985,and May 31,2025,was retrieved from the Web of Science Core Collection.Bibliometric analyses were conducted using CiteSpace 6.2.R3,VOSviewer 1.6.20,and SCImago Graphics to visualize publication characteristics,country and regional contributions,authorship patterns,journals,institutions,keywords,and cited references.Results:A total of 349 publications were included.(1)Annual publications:The number of studies on acupuncture in ophthalmology showed a generally increasing trend from 1985 to 2025.(2)Countriesegions:Research in this field spanned 33 countries and regions,with China(203 papers),the United States(55 papers),and South Korea(25 papers)as the top three contributors.(3)Institutions and authors:A total of 321 institutions and 621 authors were involved.Authors with more than three publications formed seven collaborative research teams,comprising three major collaboration networks.Xiao-peng MA was the most prolific author(12 papers),whereas Tae-Hun Kim was the most influential author(41 citations).(4)Journals:The 349 articles appeared in 152 journals,with Medicine(32 articles)and Evidence-Based Complementary and Alternative Medicine(18 articles)being the most common publication venues.(5)Keyword and reference analyses identified several major thematic areas,including dry eye syndrome,systematic reviews,etiology identification,secondary glaucoma,visual symptoms,retinal pigment research,and myopia control.Conclusion:This bibliometric analysis demonstrates that current research in acupuncture ophthalmology is primarily focused on disorders in which acupuncture shows notable clinical benefits,incorporating multimodal therapies and multidisciplinary approaches.The field is progressing toward greater protocol standardization and clearer visualization of underlying mechanisms.Acupuncture has become an important complementary and alternative therapeutic modality within ophthalmic care,offering unique advantages in symptom improvement,enhancement of quality of life,and reduction of medication dependence for conditions such as dry eye disease,asthenopia,and myopia.Future research priorities include:(1)deepening mechanistic research on acupuncture for ophthalmic diseases;(2)advancing integration with modern ophthalmology in chronic disease management and prevention;and(3)generating high-level clinical evidence to establish standardized acupuncture regimens for eye health. 展开更多

Leopards Panthera pardus are top predators that play a crucial role in maintaining ecosystem balance and stability by exerting top-down control on prey species populations through direct predation.Despite their wide distribution,in recent years leopard populations have become endangered in many regions due to increasing anthropogenic activity.Studying the feeding habits of endangered species provides insight into their survival from the perspective of their trophic niche and can help identify factors contributing to their decline,aiding in the development of targeted conservation strategies.In this study,the North China leopard(P.p.japonensis)and Amur leopard(P.p.orientalis)were selected as the study objects.A total of 97 fecal samples were collected throughout their typical habitats,83 of which were confirmed as originating from leopards using molecular identification.DNA metabarcoding identified 13 prey species for the North China leopard and 15 for the Amur leopard,with small-and medium-sized prey(5.8%)highlights the impact of anthropogenic activities on leopard feeding habits.To support leopard conservation,it is recommended to protect all prey species within reserves and reduce anthropogenic interference in these habitats. 展开更多

Fanconi anemia(FA;OMIM:227650)is a rare genetic disorder characterized by bone marrow failure,congenital anomalies,and cancer predisposition.While FANCA mutations account for most FA cases,phenotypic overlap with other disorders complicates diagnosis.This study analyzes molecular diagnostic pathways for FANCA-related FA and establishes a hereditary differential diagnosis for ectrodactyly.A Chinese FA family clinical phenotype was collected.The proband and father underwent whole-genome sequencing.Copy number variations(CNVs)in FANCA were assessed by genomic qPCR.Functional characterization of the EHMT1 variant included minigene splicing assays,RT-qPCR and Western blotting on peripheral blood samples.The proband showed pancytopenia and bone marrow hypoplasia,suggesting aplastic anemia.Sequencing analysis identified two FANCA mutations,NM_000135.4:c.154C>T and NC_000016.9:g.89865477_89895212del,which caused partial protein deletion.Subsequent pedigree analysis revealed that the affected individuals of the proband's paternal lineage,who exhibited ectrodactyly,were heterozygous for the EHMT1 c.2382+1750G>A variant(NM_024757.5)and showed significantly reduced EHMT1 mRNA expression,demonstrating complete genotype-phenotype co-segregation.Furthermore,Western blot revealed reduced H3K9me2 and decreased intensity of a~100 kDa EHMT1-reactive band in the proband's father.The newly identified g.89865477_89895212del mutation enriches the FANCA gene mutant spectrum.Additionally,the EHMT1 c.2382+1750G>A variant co-segregates with ectrodactyly and is accompanied by significantly reduced EHMT1 mRNA expression. 展开更多
